A muscle biopsy is usually performed while you are awake. Your health care provider will apply a numbing medicine (local anesthesia) to the biopsy area. During the biopsy there is usually little to no discomfort. The anesthetic may burn or sting when injected.
The physician will make a small incision, usually in your ankle or calf, and remove a small portion of your nerve. Youll likely have a few stitches at the biopsy site, but theyll be absorbed by your body and wont need to be removed.
You may feel mild discomfort when the muscle is cut. Your healthcare provider will withdraw the biopsy needle and apply firm pressure to the biopsy site for a few minutes, until the bleeding has stopped. Your provider will close the opening in your skin with adhesive strips or stitches, if needed.
You will spend about 1-1/2 hours in the Medical Procedures Unit (MPU). The muscle biopsy itself will take about 10 minutes. It will take about 45 minutes to prepare you for the test and you will spend about 30 minutes in the recovery room after the test.

The patient is instructed not to walk if the biopsied muscle is in the leg and should not use the arm to hold anything for the first 24 hours if the biopsy is done on the arm. After 24 hours and for about three to four days, light activities such as combing hair and walking inside of the home are allowed.
Wrap the clamped muscle biopsy in gauze slightly moistened with normal saline. Do not immerse in saline, as this will produce an artifact which may interfere with interpretation. Immediately deliver to University of Iowa Diagnostic Laboratories on wet ice. Package the tissue so that is stays cold, but DOES NOT FREEZE.
