Simple staining involves directly staining the bacterial cell with a positively charged dye in order to see bacterial detail, in contrast to negative staining where the bacteria remain unstained against a dark background.
The types are: 1. Simple Staining 2. Differential Staining 3. Gram Staining 4.
Basic stains include methylene blue, crystal violet, malachite green, basic fuchsin, carbolfuchsin, and safranin. Acidic stains include eosin, acid fuchsin, rose bengal, and congo red. Basic stains are attracted to negatively charged molecules in the cell including nucleic acids (DNA and RNA) and some proteins.
Microscope Slide Stains Iodine: Stains carbohydrates in plant and animal specimens brown or blue-black. ... Methylene blue: Stains acidic cell parts (like nucleus) blue. ... Eosin Y: Stains alkaline cell parts (like cytoplasm) pink. ... Toluidene blue: Stains acidic cell parts (like nucleus) dark blue.
In the histopathology laboratory, the term “routine staining” refers to the hematoxylin and eosin stain (H&E) that is used “routinely” with all tissue specimens to reveal the underlying tissue structures and conditions.
Safranin (Safranin O or basic red 2) is a biological stain used in histology and cytology. Safranin is used as a counterstain in some staining protocols, colouring cell nuclei red. This is the classic counterstain in both Gram stains and endospore staining.
How to Stain Slides Prepare a wet mount or dry mount with a coverslip. Add a small drop of stain to an edge of the coverslip. Place the edge of a tissue or paper towel on the opposite edge of the coverslip. Capillary action will pull the dye across the slide to stain the specimen.
Simple Staining 2. Differential Staining 3. Gram Staining 4. Acid Fast Staining 5.
Differential staining methods include safranin-methylene blue stain (Baxby et al., 1984), Kinyoun (Ma and Soave, 1983), Ziehl-Neelsen (Henricksen and Pohlenz, 1981), and DMSO-carbol fuchsin (Pohjola et al., 1984), which all stain the oocysts red and counterstain the background.
Note: Stain molecules can be of two types- acidic and basic. Acidic stains are utilized for background staining whereas basic stains are used for staining bacterial cells.
