Hi Im Megan today were going to stain a blood smear so that we can look at it under the microscope. Now here in this hospital we use the right stain and so the times that im going to give you for leaving the slide in each jar are consistent for what we use at this hospital - it may be a little bit different where you are. Its also important to make sure that your stain jars stay clean and that they stay free of debris. What we do here is we have a set of stain for our CBCs and cytologies, we have one for urine and we have one for fecals. And thats so that we dont cross contaminate and start to get really interesting results on our different cytologies and UAs so we make sure were not mixing things. We also empty the stain out of these jars, clean the jars and let them dry at least once a week so thats going to vary also depending on where you are and how often you use them but you can get things that grow in your stain that will show up on your blood films and its going to y

When using a stain, the usual procedure is to wet-mount the specimen, which means to place a drop of water on the slide, set the specimen in the water and cover it with a cover slip. You then apply the stain to a corner of the slide with a dropper and allow it to be drawn toward the specimen by capillary action.
The Most Common Stains Used Haematoxylin and eosin combined make up a common stain most histology labs use first. No doubt you're familiar with at least haematoxylin, which is sometimes used alone. Combining this with eosin helps bring better definition to tissues and cells using a specific color chart.
Process Prepare a wet mount slide with a specimen. Place a single drop of stain on one outer edge of the cover slip on top of your slide. Place some paper towel against the opposite edge of the cover slip - as close to the edge as possible. The paper towel will draw the stain underneath the cover slip.
