Gram staining procedure Applying a primary stain (crystal violet). Adding a mordant (Grams iodine). Rapid decolorization with ethanol, acetone or a mixture of both. Counterstaining with safranin.
Place your carefully prepared fixed smear slides on the stain rack over the sink.
To view a Gram stain: Mount the slide on the stage with the smear facing the objective lens; if the slide is upside down, you wont be able to focus at high magnification.
Put a drop of stain on an outer edge of your cover slide. Place a piece of napkin or paper towel against the opposite side of your cover slip, right up against the edge. This will help draw the stain under the cover and across the specimen.
Negative staining is a procedure, which embeds small biological particles adsorbed on an electron transparent sample support (EM grid) in a thin and amorphous film of heavy metal salts to reveal their structural details in the transmission electron microscope (Figure).
Microscope Slide Staining Information. Microscope cell staining is a technique used to enable better visualization of cells and cell parts under the microscope. By using different stains, a nucleus or a cell wall are easier to view.
Place some paper towel against the opposite edge of the cover slip - as close to the edge as possible. The paper towel will draw the stain underneath the cover slip. Wait until all of the stain has been pulled in between the cover slip and the slide. The stain should completely cover the specimen on the slide.
Simple Stain Procedure Place your carefully prepared fixed smear slides on the stain rack over the sink. Let the stain sit for 1-5 minutes. Using the clothespin, grab the long end of the slide, tilt the slide over the sink and rinse the stain off with a stream of water from the wash bottle.
The most basic reason that cells are stained is to enhance visualization of the cell or certain cellular components under a microscope. Cells may also be stained to highlight metabolic processes or to differentiate between live and dead cells in a sample.
A small square or circle of thin glass called a coverslip is placed over the specimen. It prevents the slide from drying out when its being examined. Iodine stain can be used to stain plant cells to make the internal structures more visible.
