Packing factor (PF) PF is defined as the consolidated bed height (bed height measured after settling a resin at a given flow velocity, L conc) divided by the packed bed height (Lpacked), that is, PF=L conc/ Lpacked.
Allow the slurry to settle overnight (12 hours) for best results. Determine the settled resin volume, and adjust the slurry concentration to 3050 % by adding or removing packing buffer.
The PF varies in relation to bed height and is calculated as y = 0.002x + 1.22 (where y is the calculated PF and x the packed bed height). For each bed height, the acceptable interval for the packing factor is 0.03.
A distillation column is sized by determining the diameter of the tower. An initial estimation of the tower diameter can be done based on the vapor and liquid loadings in the column. Included in this guideline is an example of the data sheet used in the industry and a calculation spreadsheet for the engineering design.
There are two common ways you can fill a chromatography column: Dry Packing. For this method, add your dry solid phase to the column and pass equilibria buffer or starting solvent to saturate the solid. Wet Packing. Here, you mix both liquid and solid outside of the column and pour it into the column.
Currently, there are three different ways to pack process chromatography columns: flow pack, which uses constant pressure or flow; dynamic axial compression, achieved by axially compressing the media slurry; and pack-in-place, which incorporates column valves into the flow cells.
6) Calculate the volume of slurry required to pack the column using the following equation; Volume 50% slurry = (Target CV x 2) x (Cf) Where Cf is the resin compression factor derived from: Cf = gravity settled bed height/flow packed bed height For example, for a 100 ml CV you will need (100 x 2) x 1.15 = 230 mL, for a
To pack a column, you must: Clean the column and packing device. Prepare the slurry solvent and packing solvent. Prepare the packing device. Prepare your LC system. Assemble the packing device and empty column. Use the backpressure regulator (if appropriate) Position the packing device.
