How to locate columns resolution

continuing on with our introduction to chromatography were going to be talking about separation and column resolution based on the number of equilibria that a mixture will undergo when we were talking in our last video we had been talking about relative retention alpha in terms of the bigger the difference between two partition ratios the better the separation if compounds have a large difference in partition ratios then theyll be easily separated with a minimum of number of equilibria but if you have two compounds that have very similar partition ratio is very similar affinities for the stationary phase its going to take a lot of equilibria to separate them therefore resolution or ability to separate things is dependent upon a ratio of their partition ratios and we call that alpha the relative retention and it is K Prime 2 thats the one that has the larger partition ratio divided by K Prime 1 so the ratio of those two partition ratios and it is always greater than 1 the bigger nu

Maintain equal distance between the centres of two columns. If using the minimum size of column 8 x 8 (200mm x 200mm) distance should not more than 3.5 m centre to centre of column distance. If the distance between the two columns is more than 3.5m than you should going for large column.
Chromatographic parameters The advantage of switching to longer columns is that the theoretical plates will increase and so will the resolution. The number of theoretical plates (N) is directly proportional to the length of the column and inversely proportional to the particle diameter2.
As a general rule, columns should be placed nearby the corners of the building, and at the points where beams and walls come together. Select the location of columns so that beams dont bend. Avoid beams that are too long. Avoid columns with a lot of space between them in the middle.
On the View menu, click Choose Details, and then click the name of the column that you want to adjust. In the Width of selected column (in pixels) box, enter the size that you want, and then click OK.
It is a well-known fact that the separation efficiency of a chromatographic column increases with an increase in its length. Doubling the column length increases resolution by around 40%.
HPLC Instrument Considerations Standard columns for reversed phase, normal phase, and ion exchange chromatography typically range from 3.9 to 4.6 mm internal diameter and 15, 25, and 30 cm in length. Standard columns for gel permeation chromatography (GPC) are 7.8 mm by 30 cm.
A longer column generally improves the separation. The trade-off is that the retention time increases proportionally to the column length and a docHub peak broadening will be observed as well because of increased longitudinal diffusion inside the column.
Well, longer columns do provide higher plate counts (more plates per column) meaning that the elution bands are sharper (reduced fraction volume) which is what you want when performing analytical HPLC. Wider columns, however, provide increased loading capacity and this is what preparative chromatography needs.
