Stochastic Optical Reconstruction Microscopy (STORM) is the one of the most widely used forms of super-resolution microscopy. Conventional microscopy is limited by the Abbes diffraction limit of 250 nm. This means that only objects larger than 250 nm can be resolved.
However, the resolution of conventional fluorescence microscopy is limited by diffraction to about 180 nm in the focal plane and to about 500 nm along the optic axis.
The fundamental principle behind PALM and related methodology is that the activated state of a photoswitchable molecule must lead to the consecutive emission of sufficient photons to enable precise localization before it enters a dark state or becomes deactivated by photobleaching.
STORM/PALM utilizes fluorescent probes that can switch between fluorescent and dark states so that in every snapshot, only a small, optically resolvable fraction of the fluorophores is detected. This enables determining their positions with high precision from the center positions of the fluorescent spots.
The main difference between PALM and STORM is the fluorophores used for the experiment and the mechanism of switching between the bright and dark states. PALM uses photo switchable/convertible fluorescent proteins (FPs), whereas STORM uses organic dyes as fluorescent probes for imaging.
The difference between PALM and fPALM is that PALM uses a TIRF microscope while fPALM uses a more traditional confocal microscope.
As STORM is a super-resolution microscopy technique, it can be used to obtain very detailed images of a sample for research and diagnostic purposes. STORM can be used to analyze the cellular structures of animal cells and has limited applications to plant cell biology.
Fluorescence microscopy is a valuable toolbox to study cellular structures and dynamics spanning scales from the single molecule to the live animal. The spatial resolution that can be achieved with any light-based microscopy is however limited to about 200 nm in the imaging plane and 500 nm along the optical axis.
