Tuberculosis skin test (or Mantoux skin test) is a test needed prior to the BCG vaccination to check it's safe to have the vaccine. This is offered to anyone aged 7 and above who has not previously received the BCG vaccine, or has no recorded history or visible BCG scar.
Administration Locate and clean injection site 5–10 cm (2–4 inches) below elbow joint. Place forearm palm-up on a firm, well-lit surface. ... Prepare syringe. Check expiry date on vial and ensure vial contains tuberculin PPD-S (5 TU/0.1 ml). ... Inject tuberculin (see Figure A3.1) ... Check injection site. ... Record information.
TB blood test (IGRA) – The TB blood test is sometimes done after or instead of the Mantoux test. If the result is negative and you have no symptoms of TB it is unlikely you have TB. If the result is positive and you have no symptoms of TB disease it is likely that you have latent TB infection.
The test is "positive" if there is a bump of a certain size where the fluid was injected. This means you probably have TB germs in your body. Most people with a positive TB skin test have latent TB infection. To be sure, your doctor will examine you and give you a chest x-ray.
A positive TB skin test will look like a firm, red bump. Your provider will measure the size of the firmness of the bump (induration). Your provider considers your particular risk factors to determine the results of your test.

Administering the TST. The Mantoux tuberculin skin test (TST) is performed by placing an intradermal injection of 0.1 ml of purified protein derivative (PPD) containing 5 tuberculin units (TU) into the volar surface of the forearm.
The Mantoux test is a widely used test for latent TB. It involves injecting a small amount of a substance called PPD tuberculin into the skin of your forearm. It's also called the tuberculin skin test (TST).
The skin test reaction should be read between 48 and 72 hours after administration. If the test is not read within 72 hours, another TST should be placed unless the amount of induration is ≥ 10 mm within 7 days after placement.
How is it done? A small amount of purified protein – taken from dead TB bacteria – is injected under the top layer of the skin, on the forearm. A sterile disposable needle and syringe is used to do this. A small blister will appear at the site, which will disappear within 20 minutes.
