Stains primarily form when a substance is spilled onto a surface and the molecules are trapped inside the fibers and pores of the material. The spilled substance coats the underlying material and the newly formed stain reflects back light of its own color, which is how the stain becomes visible to the naked eye.
Basic stains are attracted to negatively charged molecules in the cell including nucleic acids (DNA and RNA) and some proteins. Acidic stains can be positive stains (stain the cells) or they can be negative stains (stain the background and not the cells).
Microscope cell staining is a technique used to enable better visualization of cells and cell parts under the microscope. By using different stains, a nucleus or a cell wall are easier to view. Most stains can be used on non-living (fixed) cells, while only some types of stain can be used on living cells.
