Cell nuclei should be crisp, blue to black on color and the chromatin patterns of the nucleus should be well defined. Cell cytoplasm stains blue-green and keratin stains orange in color.

How do you remove Pap stains?

Procedure: Remove cover slip by incubation in xylene for a few hours to several days (until the coverslip can be removed easily). Rehydrate slides in 100%, 85%, and 75% ethanol, each for 30 sec at room temperature. Wash slides in deionized or distilled water for 5 min at room temperature.
