Papanicolaou stain (also Papanicolaous stain and Pap stain) is a multichromatic (multicolored) cytological staining technique developed by George Papanicolaou in 1942. The Papanicolaou stain is one of the most widely used stains in cytology, where it is used to aid pathologists in making a diagnosis.
Before specific staining can occur, tissue samples must undergo preparation through the following stages: Fixation, processing, embedding, sectioning, and sometimes antigen retrieval. In modern histology laboratories, most of these steps are automated.
This occurs because the stains either have a very high affinity for tissue elements and/or low affinity for processing fluids and mounting media, or at least dissolve in these latter materials very slowly.
Hematoxylin is a basic dye that is commonly used in this process and stains the nuclei giving it a bluish color while eosin (another stain dye used in histology) stains the cells nucleus giving it a pinkish stain (Victor, 2013).
Urine can be submitted in a red-top tube for cytology. Unstained, unfixed, and unoiled smears are preferred.
The slides from the cell block are usually stained by a regular Hematoxylin and Eosin (HandE) stain. These three types of stains are commonly used in different laboratories.
Youll need to take your urine cytology test after your first morning pee. This is because cells that are held in your bladder for a long period of time, or overnight, can break down over time, which can make it difficult for a pathologist to look for certain cells under a microscope.
The cells that come from the renal pelvis, ureters, bladder, and urethra are carried by the urine, and therefore, they can be observed in fresh samples and in smears with Giemsa and Papanicolaou stain.
