Start by applying oil-based stain using a Purdy brush, wiping off the excess with a lint-free rag. Then, choose a secondary tone to achieve your custom look.
Amyloid Identification Criteria It is weakly basophil with alcian blue and toluidine blue. It stains metachromatically with crystal violet and toluidine blue. It stains with congo red and sirius red F3B. Congo red and sirius red stained amyloid show green birefringence.
Congo Red and Thioflavin S are the two major histological stains used to detect any form of amyloid. These dyes bind to the characteristic -pleated sheet conformation of amyloid.
Even when the two boards are of the same type, we oftentimes discover that the same stain does not produce the same results. The cells which form the pores of an oak board, for instance, vary in their natural color from red oak, which has a natural pinkish tint, to white oak, which is actually tan.
Most commonly, amyloid can be identified with the Congo red stain as an apple-green birefringence when viewed under polarized microscopic light. Additionally, other stains may be used to identify amyloid such as crystal violet or thioflavin T.
Amyloid deposition is confirmed using thioflavin dyes, most commonly Congo red. On light microscopy, Congo red-stained amyloid deposits will appear red or salmon-pink (Fig. 1b).
Whenever leukaemia is not structurally obvious with Romanowsky stains, cytochemical stains help examine the structure of cells that are still developing. A lysosomal enzyme called myeloperoxidase (MPO) is found in the azurophilic granules of neutrophils and their precursor cells, eosinophils, and monocytes.
Blood tests to look for stress and strain on the heart are useful in many forms of heart disease, including AL amyloidosis. The cardiac biomarkers that are used include troponin T or troponin I, and NT-proBNP (which stands for N-terminal pro-brain natriuretic peptide) or BNP (brain natriuretic peptide).
