Types of Staining Based on the process, staining is grouped into three types: Simple staining, Differential staining, and Special staining. Differential staining involves multiple stains to distinguish between different types of microorganisms or structures within a specimen.
The principle of pap stain Solution 1 has Hematoxylin stain, a basic nuclear stain. Solution 2 has OG-6, an acidic cytoplasmic stain, has orange G with phosphotungstic acid (6 denotes the used concentration of phosphotungstic acid). Orange G is an acid dye and stains keratin with a bright intense orange.
Slides are thoroughly hydrated prior to hematoxylin staining. Hematoxylin solution rapidly becomes contaminated with alcohol and sometimes xylene. This causes uneven staining.
Most commonly, amyloid can be identified with the Congo red stain as an apple-green birefringence when viewed under polarized microscopic light. Additionally, other stains may be used to identify amyloid such as crystal violet or thioflavin T.
Special Stains Massons Trichrome. The trichrome stain helps to highlight the supporting collagenous stroma in sections from a variety of organs. Verhoffs Elastic Stain. Reticulin Stain. Giemsa Stain.
Composition of the Reagents of Papanicolaou Staining (Pap stain) Hematoxylin = 2.5g. Ethanol = 25ml. Potassium alum = 50g. Distilled water (50C) = 500ml. Mercuric oxide = 1-3g. Glacial acetic acid = 20ml.
The Papanicolaou stain is undoubtedly the most commonly used in cytology. It has three cytoplasmic dyes, orange G, eosin Y, and light blue. It can stain cells blue or blue-green (basophilia, cyanophilia), pink (acidophilia, eosinophilia), orange (orangeophilia), or indeterminate (gray-blue).
Special stains are used as per requirements: Modified Ziehl Neelson (for acid fast bacilli), Gram staining (Bacteria), Mucicarmine (mucins), PAS (for glycogen, fungal wall, lipofuscin, etc), Oil red O (lipids), Perls Prussian blue (iron), modified Fouchets test (bilirubin), etc.
