Put a drop of stain on an outer edge of your cover slide. Place a piece of napkin or paper towel against the opposite side of your cover slip, right up against the edge. This will help draw the stain under the cover and across the specimen.
Staining is done to visualise the specimen clearly under the microscope. Without staining the specimen under the microscope will look transparent or will not be visible clearly. Hence, staining is done to make the components of cell clearly visible.

Stains. The most widely used stains in electron microscopy are the heavy metals, uranium and lead. The double contrast method of ultrathin sections with uranyl acetate (UA) and lead citrate is the standard contrasting technique for electron microscopy (Figure 1 and 2).
The Most Common Stains Used Haematoxylin and eosin combined make up a common stain most histology labs use first. No doubt youre familiar with at least haematoxylin, which is sometimes used alone. Combining this with eosin helps bring better definition to tissues and cells using a specific color chart.
In order to stain the bacterial specimen for microscopy one must first prepare the smear on the slide. This basically involves three steps----transferring a liquid suspension of the bacterium on the slide, drying the smear, and then heating slightly to firmly attach the smear to the slide.
Osmium tetraoxide OsO4 is a widely used staining agent used in transmission electron microscopy (TEM) to provide contrast to the image. As a lipid stain, it is also useful in scanning electron microscopy (SEM) as an alternative to sputter coating.
Smear Preparation. Not only are most bacteria very small, they are also very clear and difficult to view under a microscope without first staining. You must firmly attach your bacteria to a glass slide before you can stain them.
