Inflammasomes are expressed primarily by myeloid cells and are located within the cell. The macromolecular inflammasome structure can be visualized by cryo-electron microscopy. This complex has been found to play a role in a variety of disease models in mice and several have been genetically linked to human diseases.
Prior to its activation, NLRP3 has been shown to be resident in the endoplasmic reticulum (ER) and cytosol, although, upon activation, the NLRP3 inflammasome has been shown to assemble in the cytosol, mitochondria, and mitochondria-associated ER membranes by different reports.
The alternative NLRP3 inflammasome (right) is activated in human monocytes in response to LPS and requires receptor-interacting serine/threonine-protein kinase 1 (RIPK1), FADD, and caspase-8 for its activation. This pathway is K+ efflux independent and does not induce pyroptosis.

Apoptosis-associated speck-like protein containing a CARD (ASC) has a pivotal role in inflammasome assembly and activation. While ASC function has been primarily implicated in innate immune cells, its contribution to lymphocyte biology is unclear.
It contains a bipartite structure comprising of a pyrin and a caspase recruitment domain (CARD) domain. These two domains help ASC function as an adaptor molecule. ASC is encoded by the gene PYCARD. ASC plays pivotal role in various diseases as well as different homeostatic processes.
A critical step in caspase-1 activation is the assembly of large macromolecular complex through CARD-CARD and pyrin domain–pyrin domain protein-protein interactions to form a scaffold for the recruitment and activation of procaspase-1.
Active caspase 1 is released from THP1 cells upon LPS/ATP stimulation. A) WEHD-afc cleavage in untreated or LPS/ATP treated cell lysates and supernatant (2x107 cells/ml) in the presence or absence of YVAD-cmk inhibitor (50 μM).
Caspase-1 activation by inflammasomes is a critical component of the host response to microbial pathogens. It induces secretion of HMGB1, IL-1β, and IL-18 and triggers pyroptosis of infected host cells in order to eliminate the infectious agent.
Also, enhanced NLRP3 expression can be demonstrated in SCI, predominantly in neurons, but also in microglia and astrocytes (Zendedel et al, 2016).
