Out of which, 95% ethanol is the commonly used fixative. Hence, the conventional method for fixation of Pap smear is to fix the Pap smear immediately in 95% ethyl alcohol after preparing the smear.
Smear the collected cellular material on a previously labeled glass slide. Fix immediately in 95% alcohol or with a suitable spray fixative. Note: Cellular specimens from direct lesion scrapings dry out rapidly. Thus, rapid fixation is critical if optimum results are to be obtained.
Conventional Pap Smear: Cervical scrape and brushing is recommended. Except for evaluation of maturation index and detection of vaginal adenosis, submit only one slide. For routine Pap tests, both the scraping of the squamocolumnar junction and endocervical brushing should be spread onto the same slide.
Rotate a spatula 360 once to obtain a single sample. Smear the sample onto the labeled slide. Fix the sample immediately (before it is air-dried) using a cytology spray fixative. Hold the fixative 15-20 cm (6 to 8 inches) away from the slide and evenly spray the slide by depressing the plunger 2 or 3 times.
Many people who have a Pap smear notice light bleeding, or spotting, for a couple of days after the test. In most cases, this is not a cause for concern, and it usually goes away without intervention. This symptom may occur because the Pap smear can irritate the blood vessels of the cervix, which causes light bleeding.
Smears should be fixed in alcohol or spray-fixed while wet for Pap stain. Air-dried smears are helpful for special stains and immediate evaluation of adequacy using rapid stains. It is preferable to make only two slides per aspiration attempt (one air-dried and one fixed).
It can take as long as three weeks to receive your test results. If your test shows that something might not be normal, your doctor will contact you and figure out how best to follow up. There are many reasons why test results might not be normal. It usually does not mean you have cancer.
Smears must be fixed immediately before they dry. A 95% solution of ethyl alcohol or spray fixative is used to fix the slide. The slides should be labeled in pencil with the patients name AND date of birth and a properly completed Pap smear requisition should accompany the slide.
Not use vaginal creams, foams, gels, douches or tampons 48 hours (two days) before your Pap smear. These products could cause an abnormal result. Not have sex 48 hours (two days) before your Pap smear. Sex can irritate the skin of the cervix or cause discharge (fluids) that can lead to an abnormal result.
The pap smear (slide) test is primarily used to test for cervical cancer as well as to determine if you have a bacterial or fungal infection. Your doctor would ask you to disrobe and wear a hospital gown; he would then request you to lie down on the bed, while putting your leg into stirrups.
