The pap smear (slide) test is primarily used to test for cervical cancer as well as to determine if you have a bacterial or fungal infection. Your doctor would ask you to disrobe and wear a hospital gown; he would then request you to lie down on the bed, while putting your leg into stirrups.
There are two types of tests: the Pap test and the HPV test. For both, the doctor or nurse collects cells from the surface of the cervix. With the Pap test, the lab checks the sample for cancer cells or abnormal cells that could become cancer later. With the HPV test, the lab checks for HPV infection.
Saline solution may be used for moistening. The lubricant does not dissolve during processing of the slide. It often stains dark purple and may hide numerous cells. Before scraping, the wooden spatula should be dipped in saline to moisten it and prevent too rapid dehydration of the scraped cells.
In most cases, an abnormal Pap test is a result of: A human papillomavirus (HPV) infection. A sexually transmitted infection (STI or STD), such as herpes or trichomoniasis. A bacterial or yeast infection. Inflammation in the pelvic area. Normal cellular changes that occur with age.
Pap smear results are reported as normal, abnormal, or unsatisfactory for evaluation. A normal, also called negative, Pap smear result indicates that no evidence of abnormal cells were found in the sample.
Rotate a spatula 360 once to obtain a single sample. Smear the sample onto the labeled slide. Fix the sample immediately (before it is air-dried) using a cytology spray fixative. Hold the fixative 15-20 cm (6 to 8 inches) away from the slide and evenly spray the slide by depressing the plunger 2 or 3 times.
Label the glass slide with the patients last name, first name and medical record number (or date of birth) using a graphite pencil (do not use ink, wax or crayon). Gently remove excessive secretion or blood at the cervical os with cotton swab and discard.

Conventional Pap Smear: Cervical scrape and brushing is recommended. Except for evaluation of maturation index and detection of vaginal adenosis, submit only one slide. For routine Pap tests, both the scraping of the squamocolumnar junction and endocervical brushing should be spread onto the same slide.
