Staining dyes will stain different components of the cell with different colors and intensities as follows: Nuclei: Blue. Acidophilic cells: Red. Basophilic cells: Blue Green. Erythrocytes: Orange-red to dark pink. Keratin: Orange-red. Superficial cells: Pink. Intermediate and Parabasal Cells: Blue-Green.
The principle of pap stain Solution 1 has Hematoxylin stain, a basic nuclear stain. Solution 2 has OG-6, an acidic cytoplasmic stain, has orange G with phosphotungstic acid (6 denotes the used concentration of phosphotungstic acid). Orange G is an acid dye and stains keratin with a bright intense orange.
Papanicolaou stain (also Papanicolaous stain and Pap stain) is a multichromatic (multicolored) cytological staining technique developed by George Papanicolaou in 1942. The Papanicolaou stain is one of the most widely used stains in cytology, where it is used to aid pathologists in making a diagnosis.
Pap Staining involves five dyes in three solutions, a nuclear stain and two counter stains: Nuclear Stain: Harris Haematoxylin which is used to stain cell nuclei. Counter Stain OG6: Orange G dye is used to stain keratin. Its original role was to stain the small cells of keratinizing squamous cells present in sputum.
STAINING PROCEDURE: Fix in 95% Ethyl Alcohol for 5-10 minutes. Rinse in running distilled water for 1 minute. Stain in Hematoxylin Stain, Gill I or Gill II for 2 to 6 minutes, depending on preference of nuclear stain intensity. Wash in distilled water until clear.
OG-6 is a cytologic stain routinely used in diagnostic cytology to aid in the identification and classification of exfoliative cells. OG-6 is recommended for specimens with large amounts of mucus. The stain is used for examining exfoliative cells of vaginal, cervical, sputum, as well as other body secretions.

Wash slides in deionized or distilled water for 5 min at room temperature. Destain slides in 0.5% HCl in 70% ethanol for 5-15 min at room temperature. Wash slides in deionized or distilled water for 5 min at room temperature. Air dry sections.
