Papanicolaou staining (Pap staining) is a widely used staining technique in cytopathology developed by Dr. George N. Papanicolaou. We offer a series of pap staining dyes and solutions to differentially stain various components of the cells, allowing critical examination of nuclei and cytoplasmic components.
Which are the critical points of a cytological staining? Rapid fixation in alcohol (wet version) is essential for pap staining, which brings out nuclear details clearly, allowing better identification of malignant cells.
Various fixatives are used in exfoliative cytology. Out of which, 95% ethanol is the commonly used fixative. Hence, the conventional method for fixation of Pap smear is to fix the Pap smear immediately in 95% ethyl alcohol after preparing the smear.
The principle of pap stain Solution 2 has OG-6, an acidic cytoplasmic stain, has orange G with phosphotungstic acid (6 denotes the used concentration of phosphotungstic acid). Orange G is an acid dye and stains keratin with a bright intense orange.
The PAP stain is a polychromatic counterstaining method consisting of stains such as Orange G 6 (OG6) and modified eosin azure (EA). The strength of the Pap stain is such that it results in: Well-stained nuclear chromatin. Differential polychromatic counterstaining of cytoplasm.
The most commonly used fixatives for diagnostic pathology and cytologic specimens are 10% NBF and 95% ethanol [3]. In our institution, 96% alcohol used by clinicians as a fixative for cytology specimens because of its accessibility and affordability.
