Acceptable specimens for the Pap Test include cervical, cervical and vaginal, endocervical, vaginal and/or vulvar scrapings or brushings.
Pap test. In a Pap test, your doctor uses a vaginal speculum to hold your vaginal walls apart and to see the cervix. Next, a sample of cells from your cervix is collected using a small cone-shaped brush and a tiny plastic spatula (1 and 2).
The phrase endocervical cells present simply means that your doctor sampled cells from the inside of your cervix during the Pap smear. The phrase squamous metaplastic cells present means that the pathologist who examined your Pap smear found cells that were growing and repairing themselves regularly.
Because the Papanicolaou stain was developed to document keratinization (the orange G dye component binds avidly to keratin), it is supe- rior to Romanowsky-type stains for this purpose. Identification of cytoplasmic keratinization may be essential for diagnosing squamous cell carcinomas.
The modified PAP stain showed a positive staining of KP in 30 (78.9%) cases, whereas HE stain showed positive staining in 20(52.6%) cases (Table-1, Graph-1).
Your healthcare provider will use a small brush, swab or spatula to gently remove cells from the cervix and back of the vagina. He or she will place the cells in a vial of liquid or smear the cells on a glass microscope slide. If you need an HPV test, your provider will take a sample of cells for this test as well.
The PAP stain is a polychromatic counterstaining method consisting of stains such as Orange G 6 (OG6) and modified eosin azure (EA). The strength of the Pap stain is such that it results in: Well-stained nuclear chromatin. Differential polychromatic counterstaining of cytoplasm.

3. Low-grade squamous intraepithelial lesion (previously Class III, mild dysplasia): This classification is for abnormal cells, which may be considered as mild dysplasia or with mild premalignant potential. This same category would be used if there is any sign of the human papilloma (wart) virus.
