A folder with a flashing question mark means that your computers startup disk is no longer available or doesnt contain a working Mac operating system.
Its very easy to get rid of that question mark. Right-click on it, select Options and then Remove From Dock, as shown below.

Heres how to do that. Boot your Mac into recovery mode. In macOS Utilities, select Disk Utility. Select your startup disk in the sidebar and click First Aid. Once Disk First Aid has finished repairing your disk, restart normally.
