What is the capsule mordant? The capsule mordant contains a stain called acid fuchsin. The acid pH of the mordant changes Congo red to a blue color. What does the acid fuchsin in the capsule mordant do? The acid fuchsin stains the bacterial cells a pink color.
The counterstaining is provided by Congo red. In the procedure for Manevals capsule staining method, the bacterial cells are first introduced in to a drop of Congo red solution on a slide. Congo red is a pH indicator that is blue at or below pH 3.0 and red above pH 5.2.
In general, counterstains are designed to provide a contrast of nucleus with cytoplasmic immunostaining and surrounding tissue.
Explanation: Gram staining is a type of differential staining. In this process the fixed bacterial smear is subjected to the following staining reagents in the order listed: crystal violet, iodine solution, alcohol (decolorizing agent), and safranin.
The primary stain applied is crystal violet, which stains both the bacterial cell and the surrounding capsule. A 20% copper sulfate solution is then applied, which serves a dual function as both decolorizer and counterstain. It removes and replaces the crystal violet in the capsule only.
In Anthonys capsule stain, crystal violet is used as the primary stain, interacting with the protein material in the culture broth or added during the staining, and copper sulfate serves as the mordant.
Without the crystal violet staining, it would be impossible to differentiate between Gram-positive and Gram-negative bacteria, as the secondary counterstain, usually safranin, would stain all cells pink instead of just the decolorized Gram-negative cells.
Acid-Fast Stains Both use carbolfuchsin as the primary stain. The waxy, acid-fast cells retain the carbolfuchsin even after a decolorizing agent (an acid-alcohol solution) is applied. A secondary counterstain, methylene blue, is then applied, which renders nonacid-fast cells blue.
