- The cervix is the lower narrow end of the uterus that connects to the top of the vagina. The cervix is covered by a thin layer of tissue made up of cells. Cancer of the cervix, or cervical cancer, can develop when cells become abnormal. Most cases of cervical cancer are caused by infection with a virus called HPV, or human papillomavirus. The HPV virus can cause cells to become abnormal over time. One way to prevent HPV infection is by getting the HPV vaccine. You can also prevent cervical cancer by having regular screening tests and follow-up treatments if needed. Cervical cancer screening may include Pap tests, HPV tests, or both. Pap tests check for abnormal cells in your cervix. HPV tests check for the HPV virus. In both tests, cells are taken from the cervix in the same way and sent to a lab for testing. Typically, we first think of Pap tests (Pap smears) as a specific type of cytological test identifying cancerous cells in cervical tissue. However, Pap stains can also be conducted on non-gynecological specimens from a variety of bodily secretions and from small needle biopsies of organs and tissues.
Wash slides in deionized or distilled water for 5 min at room temperature. Destain slides in 0.5% HCl in 70% ethanol for 5-15 min at room temperature. Wash slides in deionized or distilled water for 5 min at room temperature. Air dry sections.
Pap Staining involves five dyes in three solutions, a nuclear stain and two counter stains: Nuclear Stain: Harris Haematoxylin which is used to stain cell nuclei. Counter Stain OG6: Orange G dye is used to stain keratin. Its original role was to stain the small cells of keratinizing squamous cells present in sputum.
Saline solution may be used for moistening. The lubricant does not dissolve during processing of the slide. It often stains dark purple and may hide numerous cells. Before scraping, the wooden spatula should be dipped in saline to moisten it and prevent too rapid dehydration of the scraped cells.
Ethyl alcohol is the fixative specifically recommended for cytological preparations. Fixation coarsens the cell structures and sharpens nuclear chromatin pattern and its details.
Following an abnormal Pap smear, your next step is additional testing. Dr. Tajzoy may suggest redoing the Pap smear to see if results come back the same. In addition to a retest, you can expect a thorough evaluation of your overall health.
The conventional Pap stain has 24 solutions, of which three are acid dyes, six water, one is chemically defined bluing agent, eight 95% ethanol, three absolute alcohol and three xylene Gill, 2006. The cytomorphology with conventional Pap stain is shown in Figure 2 .
