NMDA receptors are commonly thought to play a role in the development of cortical circuitry, primarily as mediators of activity-dependent plasticity (Kirkwood and Bear, 1994;Katz and Shatz, 1996). AMPA receptors are commonly thought to play a role in normal, ongoing transmission between neurons.
AMPA receptors undergo fast, submillisecond activation by glutamate, leading to depolarization of the postsynaptic membrane and relief of magnesium block of NMDA receptors, followed by rapid desensitization and deactivation on a millisecond time scale (1).
NMDA receptors contribute prominently to responses in the preferred direction, and their contribution to responses in the nonpreferred direction is reduced docHubly by GABAergic inhibition. AMPA receptors decrease spatial phase-selective simple cell responses and generate phase-invariant complex cell responses.
4.20. AMPA receptors of course provide the initial depolarization, either locally or distally in the neuron, that ultimately results in NMDA receptor activation. As such, alterations in the AMPA receptor protein or its associated interacting proteins can lead to loss of proper regulation of NMDA receptor activation.
In response to a brief pulse of glutamate, mimicking synaptic release, NMDARs activate slowly over hundreds of milliseconds and continue activating long after all glutamate has been removed from the synaptic cleft, thereby briefly memorizing the occurrence of a synaptic input.
NMDA receptors play an important role in the strengthening of synapses, which is known as long-term potentiation (LTP). It is an important neurological process associated with strong memory formation. NMDA receptors are involved in LTP in many brain regions, especially the hippocampal CA1 region.
NMDA (N-methyl-D-aspartate) receptor-dependent long-term potentiation (LTP) and long-term depression (LTD) of signal transmission in excitatory neurons, such as hippocampal pyramidal neurons, is thought to underlie the formation of neuronal circuits during learning and memory1,2,3.
We thus conclude that LTP induction in CA1 pyramidal neurones leads to an increase in the extrasynaptic pool of AMPA receptors in dendritic regions near activated synapses.
