The main reason you stain a specimen before putting it under the microscope is to get a better look at it, but staining does much more than simply highlight the outlines of cells. Some stains can penetrate cell walls and highlight cell components, and this can help scientists visualize metabolic processes.
Bacteria appear opaque when observed unstained under a microscope. Staining is required to make the cells and their internal structures more visible under the light microscope by increasing the contrast between the organism and the background.
Microscope cell staining is a technique used to enable better visualization of cells and cell parts under the microscope. By using different stains, a nucleus or a cell wall are easier to view. Most stains can be used on non-living (fixed) cells, while only some types of stain can be used on living cells.
Stain the fence from top to bottom with small, quick brush strokes rather than huge ones for even coverage. Its similar to deck staining in the fact that you should absolutely never start or stop in the middle. For best results, do two coats both on the same day.
Stain the Wood Thoroughly stir the wood stain before applying. Use a rag or cloth instead of a sponge, which may absorb the stain. When applying, the cloth should be wet but not dripping. You can apply stain across the grain of the wood or with the grain of the wood. Stain usually cannot be removed after application.
The most basic reason that cells are stained is to enhance visualization of the cell or certain cellular components under a microscope. Cells may also be stained to highlight metabolic processes or to differentiate between live and dead cells in a sample.
Stain can be applied with a bristle brush, a foam brush, or a cloth. On woods with large, open pores, such as oak, mahogany and ash, increase your pressure to work the stain into the pores. Rubbing or brushing against the direction of the grain will help fill deep pores with stain.
The main purpose of staining is to highlight the specimen by increasing the contrast between the organisms and the background. Some common examples of stains are methylene blue, crystal violet, carbon fuchsin, etc. Hence, staining improves the observations of any particular cell or tissue when seen under a microscope.
