The positive and negative controls are bacterial smears we use to test if the Gram stain was performed properly. If the positive and negative controls are not as expected, you cannot trust that the stain was performed properly. As a Positive Control we use a known Gram-positive bacteria.
Negative staining protocols. The conventional negative staining protocol involves the adsorption of the specimen to a glow-discharged carbon-coated EM grid, which is washed with two drops of deionized water and subsequently stained with two drops of heavy metal solution.
Then, a solvent is added to the stain. When the solvent combines with bacteria in a sample, the bacteria will either stay purple or turn pink or red. If the bacteria remain purple, they are Gram-positive. If the bacteria turn pink or red, they are Gram-negative.

For negative staining the sample is placed on a support film, bathed in a solution of a heavy metal salt that is subsequently blotted, and the sample is then left to dry in air.
Negative staining employs the use of an acidic stain and, due to repulsion between the negative charges of the stain and the bacterial surface, the dye will not penetrate the cell. In negative staining, the results yield a clear cell with a dark background.
