In contrast to direct stains that bind to bacteria directly, a negative stain colors the background of a smear rather than the bacteria. These stains have negatively charged functional groups so they cant bind directly to negatively charged bacteria.
Negative staining is performed for the observation of intact microbial structures without disturbing its cellular morphology. It is referred to as negative staining because the bacteria cells are not stained, rather the glass background containing cells.

Negative staining allows visualization of bacterial cell morphology without directly staining the cells. It works by using acidic stains like India ink or nigrosin that stain the background glass slide rather than the negatively charged bacterial cells.
The structures which can be negatively stained are much smaller than those studied with the light microscope. Here, the method is used to view viruses, bacteria, bacterial flagella, biological membrane structures and proteins or protein aggregates, which all have a low electron-scattering power.
Some suitable negative stains include ammonium molybdate, uranyl acetate, uranyl formate, phosphotungstic acid, osmium tetroxide, osmium ferricyanide and auroglucothionate. These have been chosen because they scatter electrons strongly and also adsorb to biological matter well.
Alternatively, positive and negative staining techniques can be combined to visualize capsules: The positive stain colors the body of the cell, and the negative stain colors the background but not the capsule, leaving halo around each cell.
