Negative Staining Place a drop of the diluted material on a formvar and carbon covered grid. After 30-60 seconds, blot off the excess liquid with filter paper. Add a drop of PTA solution. After 1 minute, blot off the excess with filter paper. Allow the grid to air dry. Examine in the transmission electron microscope.
It is a progressive and differential stain that generally uses 5 dyes in 3 solutions. The color reaction depends on the type of dyes used in the Papanicolaou stain. It uses hematoxylin as nuclear stain, with 2 counterstains (orange G and Eosin Azure or EA).
The conventional Pap stain has 24 solutions, of which three are acid dyes, six water, one is chemically defined bluing agent, eight 95% ethanol, three absolute alcohol and three xylene Gill, 2006. The cytomorphology with conventional Pap stain is shown in Figure 2 .
The Papanicolaou stain is undoubtedly the most commonly used in cytology. It has three cytoplasmic dyes, orange G, eosin Y, and light blue.
Pap Staining involves five dyes in three solutions, a nuclear stain and two counter stains: Nuclear Stain: Harris Haematoxylin which is used to stain cell nuclei. Counter Stain OG6: Orange G dye is used to stain keratin. Its original role was to stain the small cells of keratinizing squamous cells present in sputum.
PAP bluing solution is a variant of Scotts solution and is used specifically in Papanicolaou staining (Pap staining) for cytological smears. It is used to bluish the hematoxylin in the stained nuclei to improve contrast and visibility, which facilitates the interpretation and analysis of cytology specimens.
Wash slides in deionized or distilled water for 5 min at room temperature. Destain slides in 0.5% HCl in 70% ethanol for 5-15 min at room temperature. Wash slides in deionized or distilled water for 5 min at room temperature. Air dry sections.
