Commonly used negative stains include uranyl acetate (UA), uranyl formate, ammonium molybdate, and phosphotungstic acid. The technique was invented by Brenner and Horne (1959) and first applied on mosaic viruses mixed with phosphotungstic acid.
Some suitable negative stains include ammonium molybdate, uranyl acetate, uranyl formate, phosphotungstic acid, osmium tetroxide, osmium ferricyanide and auroglucothionate. These have been chosen because they scatter electrons strongly and also adsorb to biological matter well.
Negative staining protocols. The conventional negative staining protocol involves the adsorption of the specimen to a glow-discharged carbon-coated EM grid, which is washed with two drops of deionized water and subsequently stained with two drops of heavy metal solution.
Types of Staining Based on the process, staining is grouped into three types: Simple staining, Differential staining, and Special staining. Differential staining involves multiple stains to distinguish between different types of microorganisms or structures within a specimen.
Some stains commonly used for simple staining include crystal violet, safranin, and methylene blue.
Special Stains Massons Trichrome. The trichrome stain helps to highlight the supporting collagenous stroma in sections from a variety of organs. Verhoffs Elastic Stain. Reticulin Stain. Giemsa Stain.
Negative staining is a procedure which embeds small biological molecules such as proteins, protein complexes, or nanoparticles on a TEM grid in a thin film of heavy metal salts (i.e. uranyl salts) to enhance contrast and reveal their structural details. Unlike cryo-TEM, this process is carried out at room temperature.
Negative-staining with nigrosin or India ink is a quick and easy method to gain information about the presence or absence of capsules or any other layers around bacteria. An organism with a capsule will show a halo around the cell.
