The most commonly used stains are uranyl acetate (pH 4), ammonium molybdate (pH 57), methylamine tungstate (pH 6), and phosphotungstic acid (pH 58).
Negative staining methods make use of acid dyes such as India ink and nigrosin (a mixture of black synthetic dyes). Since these dyes are negatively charged, they are repelled by the negative charge of bacterial surfaces and do not penetrate the cells.
Negative staining requires the use of acid stain such as the Indian ink or nigrosin. The acid stain with its negatively charged chromogen will not penetrate the cells because of the negative charge on the surface of the bacteria.
Negative staining protocols. The conventional negative staining protocol involves the adsorption of the specimen to a glow-discharged carbon-coated EM grid, which is washed with two drops of deionized water and subsequently stained with two drops of heavy metal solution.
Electron microscopy is a useful tool to examine virus morphology. A negative staining technique uses heavy metal salts to enhance the contrast between the background and the virions image. This is a very simple and direct technique. Pelleted viral particles are resuspended in distilled water.
Procedure of Negative Staining Place a slide against the drop of suspended organisms at a 45 angle and allow the drop to spread along the edge of the applied slide. Push the slide away from the drop of suspended organisms to form a thin smear. Air-dry. Note: Do not heat fix the slide.
URANYL ACETATE A 1% to 3% solution of uranyl acetate dissolved in distilled water (pH 4.2 to 4.5) can be used to negatively stain many types of samples. The stain should be filtered through a 0.22 m filter that has been pre-rinsed with large volumes of double distilled water.
