Bacteria are identified routinely by morphological and biochemical tests, supplemented as needed by specialized tests such as serotyping and antibiotic inhibition patterns. Newer molecular techniques permit species to be identified by their genetic sequences, sometimes directly from the clinical specimen.
Gram-positive bacteria are bacteria classified by the color they turn in the staining method. Hans Christian Gram developed the staining method in 1884. The staining method uses crystal violet dye, which is retained by the thick peptidoglycan cell wall found in gram-positive organisms.
Under a microscope, gram-positive bacteria appear purple-blue because their thick peptidoglycan membrane can hold the dye. The bacteria is called gram-positive due to the positive result. Gram-negative bacteria stain pink-red. Their peptidoglycan layer is thinner, so it doesnt retain the blue color.
The gram-positive bacteria retain the crystal violet colour and stain purple whereas the gram-negative bacteria lose crystal violet and stain red. Thus, the two types of bacteria are distinguished by gram staining. Gram-negative bacteria are more resistant to antibodies because their cell wall is impenetrable.
Gram-negative organisms associated with lowering of milk quality can be placed into two groups, coliforms and noncoliforms. Coliforms include Citrobacter spp., Enterobacter spp., Escherichia coli, and Klebsiella spp. Legal limits for coliform counts, unlike for pasteurized milk, have not been established for BTM.
A Gram stain is colored purple. When the stain combines with bacteria in a sample, the bacteria will either stay purple or turn pink or red. If the bacteria stays purple, they are Gram-positive. If the bacteria turns pink or red, they are Gram-negative.
