Any basic dyes, such as methylene blue, crystal violet, malachite green, or safranin work well.
First cells are stained with crystal violet, followed by the addition of a setting agent for the stain (iodine). Then alcohol is applied, which selectively removes the stain from only the Gram negative cells. Finally, a secondary stain, safranin, is added, which counterstains the decolorized cells pink.
Burkes modification of the Gram Stain adds sodium bicarbonate to the crystal violet solution. Sodium bicarbonate prevents the acidification of the solution as iodine oxidizes (McClelland, 2001) and uses an aqueous solution of Safranin for the counterstain (Gephardt et al., 1981).
Gram staining is a common technique used to differentiate two large groups of bacteria based on their different cell wall constituents. The Gram stain procedure distinguishes between Gram positive and Gram negative groups by colouring these cells red or violet.
A Gram stain will group the bacteria by color and shape to help determine which kind of bacteria is causing your infection. There are two main categories of bacterial infections: Gram-positive and Gram-negative. The categories are determined by how the bacteria react to the Gram stain test.
The final step in gram staining is to use a basic fuchsin stain to give decolorized gram-negative bacteria a pink color for easier identification. It is also known as counterstain. Some laboratories use safranin as a counterstain; however, basic fuchsin stains gram-negative organisms more intensely than safranin.
A Gram stain helps diagnose harmful bacteria. Under a Gram stain, different kinds of bacteria change one of two sets of colors (pink to red or purple to blue) under a special series of stains and are categorized as gram-negative or gram-positive, ingly.
Without the crystal violet staining, it would be impossible to differentiate between Gram-positive and Gram-negative bacteria, as the secondary counterstain, usually safranin, would stain all cells pink instead of just the decolorized Gram-negative cells.
