Morgan began breeding the white-eyed mutant fly and found that in one generation of flies, the trait was only present in males. Through more breeding analysis, Morgan found that the genetic factor controlling eye color in the flies was on the same chromosome that determined sex.
Linkage analysis. A method of mapping genes using fam studies to determine if 2 genes are linked. Estimates location of disease allele through co-segregation of disease and markers within families (i.e. how often are the marker allele and disease transmitted from parent to child.
Learn about Thomas Hunt Morgan, the first person to definitively link trait inheritance to a specific chromosome and his white-eyed flies. One day in 1910, American geneticist Thomas Hunt Morgan peered through a hand lens at a male fruit fly, and he noticed it didn't look right.
A gene-hunting technique that traces patterns of disease in high-risk families. It attempts to locate a disease-causing gene by identifying genetic markers of known chromosomal location that are co-inherited with the trait of interest.
In some instances, such as for autosomal recessive and X-linked recessive traits, grandparents do not help to set the phase because they are usually unaffected and disease allele carriers cannot be distinguished from non-carriers.
1.that the inheritance of each trait is determined by "units" or "factors" that are passed on to descendents unchanged (these units are now called genes )2.that an individual inherits one such unit from each parent for each trait1 more row
Linkage studies have a number of intrinsic problems including: (1) Non-paternity, which will give a false connection between a DNA polymorphism and the disease gene being studied, and (2) Recombination of DNA segments – which is a function of the distance between a polymorphic marker and the gene of interest.
Therefore, the number of linkage groups present in an individual corresponds to number of chromosomes in its one genome (all the chromosomes if haploid or homologous pairs if diploid). It is known as principle of limitation of linkage groups. The size of the linkage group depends upon the size of chromosome.
The primary difference between these two approaches is that linkage analysis looks at the relation between the transmission of a locus and the disease/trait within families, whereas association analysis focuses on the relation between a specific allele and the disease/trait within population.
Alleles positioned on the same chromosome are not always inherited together because during meiosis linked genes can became unlinked. Frans Janssen suggested chromosomes become unlinked during homologous recombination, a process where homologous chromosomes exchange segments of DNA.
