friends today we discuss about the straining techniques we know the live bacteria do not show much structural detail under the light microscope due to the lack of contrast hence it is very customary to use training techniques to produce a color contrast the bacteria may be stained in the living state or by killing them routinely we use methods of straining of bacteria that involve drying and fixing the smear which kill the bacteria okay then uh the protoplasm of the bacteria have an Astic nature so the bacteria always have an affinity to basic ties the staining techniques that commonly used in bacteriology are simple stains negative staining impregnation methods and differential stains and simple stains a we use a methylene blow or a basic Fusion they provide a color contrast but they embed same color to all the bacteria so that we cannot differentiate between various types of bacteria and a negative staining uh we use enthane ink or nigrosin that are emulsifier with the sample or orga

The correct sequence of basic steps for capsule staining is as follows: prepare a thin wear by dragging a clean slide through the sample, allow the sample to thoroughly air dry, apply primary stain (India ink or nigrosin) to a clean slide, use an inoculating loop to sceptically mix the bacterial sample with the primary
Staining Procedure Deparaffinize sections with xylene-Peanut oil or xylene-mineral oil solution: 2 changes, 6 minutes each. Rinse with running tap water for 1 minute. Stain with Carbol Fuchsin for 20 minutes at room temperature. Wash in running tap water for 3 minutes.
What is the correct order of staining reagents in Gram-Staining? Explanation: Gram staining is a type of differential staining. In this process the fixed bacterial smear is subjected to the following staining reagents in the order listed: crystal violet, iodine solution, alcohol (decolorizing agent), and safranin.
Laboratory staff and scientists use different mounting techniques, combined with dyes and microbiology stains to add contrast to specimens and make them easier to observe at a microscopic level and aid with identification. This is what is known as staining in microbiology.
Congo Red Capsule Stain Procedure Place a loop-full of Congo Red on a slide. Mix a small amount of your organism into the drop of Congo Red. Let the slide thoroughly air dry. Fix the dried slide with acid alcohol for 15 seconds. Rinse with distilled water and cover the slide with acid fuchsin for 1-5 minutes.
Basic Steps of capsule staining: Apply primary stain (India ink or nigrosin) to a clean slide. use inoculating loop to aseptically mix bacterial sample with primary stain. create a thin smear by dragging a clean slide through the sample. allow sample to thoroughly dry. apply secondary stain (crystal violet) to smear.
The final step in gram staining is to use a basic fuchsin stain to give decolorized gram-negative bacteria a pink color for easier identification. It is also known as counterstain. Some laboratories use safranin as a counterstain; however, basic fuchsin stains gram-negative organisms more intensely than safranin.
PROCEDURE (to be done individually) Stir up the Skim Milk broth culture with your loop and place 2-3 loops of Klebsiella aerogenes on a microscope slide. Using your inoculating loop, spread the sample out to cover about one inch of the slide. Let it completely air dry. Stain with crystal violet for one minute.
