H and E staining helps identify different types of cells and tissues and provides important information about the pattern, shape, and structure of cells in a tissue sample. It is used to help diagnose diseases, such as cancer. Also called hematoxylin and eosin staining.
Clinically, we de-stain HE slides as a regular protocol using 1:1 Xylene/Acetone solution to remove the coverslip and 1% Acid (HCl) Alcohol to remove the stains. Acid alcohol is used in regressive hematoxylin staining regularly and eosin is quickly removed by both acetone and alcohol.
Eosin shows the cytoplasm, collagen, docHub tissue, and other structures that surround and support the cell as an orange-pink-red color.
Protocol for HE staining: Place a drop of Permount on the slide using a glass rod, taking care to leave no bubbles. Angle the coverslip and let fall gently onto the slide. Allow the Permount to spread beneath the coverslip, covering all the tissue. Dry overnight in the hood.
Many HE and special stain protocols begin with step #1 as simply Deparaffinize and hydrate to water. In fact, nothing could be more complicated. The sections to be stained are surrounded by a matrix of paraffin wax, which is insoluble in water. All of the paraffin must be removed to ensure optimal staining.
As the paraffin wax is insoluble in water, it is necessary to remove it from the tissue section before applying any aqueous or water-based dye solution, to allow the tissue to successfully interact with the stain.
Differentiation is a procedural staining step related to a regressive HE staining method. When staining with hematoxylin, hydrochloric acid (for rapid differentiation) and acetic acid (for slower, more controlled differentiation) are the most commonly used components of differentiating solutions.
One of the steps in the HE procedure is bluing. As the name implies, this step converts the initial soluble red color of the hematoxylin within the nucleus to an insoluble blue color. The alkaline pH of the bluing solution causes the mordant dye-lake to reform in the tissue and become more permanent.
