Flow cytometry protocol Stage 1 - Sample preparation. Stage 2 - Live/dead staining with a viability dye. Stage 3 - Fixation and permeabilization (optional - only for intracellular staining) Stage 4 - Blocking. Stage 5 - Antibody incubation. Stage 6 - Detection and data analysis.
The detection of intracellular cytokines is a relatively new technique in which cells are permeabilized and antibodies are used to detect cytokine protein within cells by flow cytometry (17, 19, 20).
While cell surface proteins, such as the CD (cluster of differentiation) antigens, are widely used for characterizing different cell types by flow cytometry, detecting intracellular molecules can often provide a more dynamic picture, says Adam Best, Ph. D., Sr. Global Marketing Manager at BD.
Intracellular staining For intracellular staining, we add the antibodies to 0.1% Tween in PBS/2% FBS. Stain 106 cells in 100 l buffer. The Ab concentration will vary, depending on the Ab. Incubate 30 min on ice.
Flow cytometry can be used to detect fluorescent antibody-tagged proteins, nucleic acids, and other subcellular components, fluorescent and fluorescently-tagged proteins, nuclear stains, dyes that measure cell membrane permeability and cell viability, and more.
Flow cytometry can be used to analyze various intracellular molecules including phosphorylated signaling proteins and cytokines.
Except where noted in the protocol, all staining should be done on ice or at 4C with minimal exposure to light. The fixation and permeabilization steps that are required for the detection of intracellular antigens may alter the light scatter properties of cells and may increase non-specific background staining.
