introducing the sample into a high pressure flow path is quite a challenge the technique that we use in hplc is to First Park the sample in a sample loop as this can be carried out just at ambient pressure after this the flow is redirected through the valve and the sample is transferred out of the loop and onto the column this animation shows a typical six Port valve contained in a manual injector the sample is introduced into the sample Loop and this is often done just with a manual syringe and then the sample Loop is completely filled therefore to vary the injection volume the sample Loop itself would have to be physically changed
