A liquid sample is injected into a stream of solvent (mobile phase) flowing through a column packed with a separation medium (stationary phase). Sample components separate from one another by a process of differential migration as they flow through the column.
Chromatographic Technique: Gas Chromatography (GC) The injector port is a hot chamber containing a glass liner where the compounds are volatilized and swept by a stream of inert carrier gas into the column.
Generally speaking, two injection systems will be available, either a manual injection, or an automatic one. In HPLC, whether the injection is manual or automatic, the injection is performed with an injection loop that must be filled and then injected into the system.
The injection is an important step in chromatography. As its name suggests, the aim is to inject the sample to be analyzed into the HPLC system. Generally speaking, two injection systems will be available, either a manual injection, or an automatic one.
An HPLC injector allows the introduction of a precise sample volume onto the column. A typical manual injector consists of a 6-port valve with a rotor, a sample loop and a needle port (Figure 9). A sample solution is introduced into the sample loop using a 22-gauge blunt tip syringe in the LOAD position.
The most common injection method is where a microsyringe is used to inject sample through a rubber septum into a flash port at the top of the column. The temperature of the sample port is usually about 50C higher than the boiling point of the least volatile component of the sample.
PROCEDURE Weigh the sample into a syringe. Confirm that the lever is in the INJECT position, insert the syringe all the way into the injection port, and quickly switch the valve to the LOAD position. Press the Inject button in Empower to wait for injection.
The simplest way to inject your sample into the system. Use a manual injection valve and choose from a large range of different sample loops, available for different applications. For analytical HPLC, our valves feature a wide back pressure range of up to 1200 bar with a 0.3 mm bore size.
