hello this is scott Kephart with quasar instruments and today iamp;#39;m going to talk about prepard of HPLC injection volumes to start out Iamp;#39;m going to draw a typical two position six port injection valve over here and do my other videos I go how to plumb this this type of injection port up but in this one weamp;#39;re just gonna be talking about the appropriate injection volume amount so what Iamp;#39;m doing is drawing the sample loop here in this case this will be from your injector this will be waste this will be from your pump and this will be to your column so what weamp;#39;re going to do is really look at the injection loop so Iamp;#39;m gonna draw that injection loop out as a straight tube and then Iamp;#39;m gonna draw vertical lines to represent the injection volume so there are the total injection amount so this is actually your injection loop and right here and right here is where your injection loop is plugging into the valve now when you load the sample on

Answer and Explanation: In the gas chromatography, to get a sharp and resolved band the amount of sample must be injected at the same time. The large amount of sample or the extra time to inject the sample in GC leads to the poor band.
The sample is introduced into the injection port at a lower temperature, and then the temperature is rapidly increased to vaporize the sample and transfer it into the column. PTV is particularly useful for analyzing samples with a wide range of boiling points. It is important to mark the solvent front immediately in paper chromatography to know how far the solvent traveled up the paper and to see the separation of the different components in the mixture. 2. For packed columns, sample size ranges from tenths of a microliter up to 20 microliters. Capillary columns, on the other hand, need much less sample, typically around 10-3 L, split/splitless injection. Purpose of port is to flash evaporate your sample and introduce it into the column.

The shorter the length of the column occupied by the injected sample, the shorter the band as it begins and completes the chromatographic process. The injection is an important step in chromatography. As its name suggests, the aim is to inject the sample to be analyzed into the HPLC system. Generally speaking, two injection systems will be available, either a manual injection, or an automatic one.
Column overloading in chromatography occurs when we inject too much sample onto the column. Injecting too much sample volume corresponds to volume overload and will result in peak broadening. Peaks can either stay symmetrical or exhibit tailing. Volume overload is hard to demonstrate, and it is fairly rare. The simplest way to inject your sample into the system. Use a manual injection valve and choose from a large range of different sample loops, available for different applications. For analytical HPLC, our valves feature a wide back pressure range of up to 1200 bar with a 0.3 mm bore size.
