pressure balance sampling is actually was developed in the late 60s it was a revolutionary technique in order to transfer volatile materials it was designed for blood alcohol specifically and was able to transfer that in a highly precise manner to a gas chromatograph pressure balance sampling works very simply what we do is we have a vial and in that vial we put a neater and pressurize it to approximately the head pressure of the column we then close the valves and it actually sends an aliquot over to the column and then when we turn that off it sends a very pressurized slug of our analyte right onto our column given an outstanding performance the pressure balance sampling Works in a variety of different application areas virtually most of them are all well suited to headspace analysis so could be looking at volatile materials and drugs looking at the off layers in beer and wine looking at food packaging of course we want our food to be safe and we want to make sure that this p

The most common injection method is where a microsyringe is used to inject sample through a rubber septum into a flash port at the top of the column. The temperature of the sample port is usually about 50C higher than the boiling point of the least volatile component of the sample.
With on-column injection a liquid sample is introduced directly into the column with a thin injection needle. During the course of the temperature program the vapour pressure of the solutes increases and the chromatographic process begins. With this injection technique no evaporation in a heated space takes place.
Headspace sampling: In this technique, the liquid or solid sample is added to a glass vial until it establishes equilibrium. Some of the analytes vaporize from the liquid or solid and enter the headspace above the sample.
The injection is an important step in chromatography. As its name suggests, the aim is to inject the sample to be analyzed into the HPLC system. Generally speaking, two injection systems will be available, either a manual injection, or an automatic one.
A micro syringe injects sample through a rubber septum into a Flash vaporiser. The temperature of the sample port is usually about 50C higher than the boiling point of the lowest boiling component of the sample mixture. The injection volumes generally range between 0.5 to 5l.
