The injection is an important step in chromatography. As its name suggests, the aim is to inject the sample to be analyzed into the HPLC system. Generally speaking, two injection systems will be available, either a manual injection, or an automatic one.
Sample injection port The most common injection method is where a microsyringe is used to inject sample through a rubber septum into a flash vapouriser port at the head of the column. The temperature of the sample port is usually about 50C higher than the boiling point of the least volatile component of the sample.
Answer and Explanation: In the gas chromatography, to get a sharp and resolved band the amount of sample must be injected at the same time. The large amount of sample or the extra time to inject the sample in GC leads to the poor band.
