Saline solution may be used for moistening. The lubricant does not dissolve during processing of the slide. It often stains dark purple and may hide numerous cells. Before scraping, the wooden spatula should be dipped in saline to moisten it and prevent too rapid dehydration of the scraped cells.
Pap test. During a Pap test, a tool called a speculum holds the vaginal walls apart. A sample of cells from the cervix is collected using a soft brush and a flat scraping device called a spatula (1 and 2). The cells are placed in a bottle that contains a solution to preserve them (3).
A speculum is a thin piece of plastic or metal with a hinged piece on one end that allows it to open and close. If the speculum is metal, the doctor or nurse will warm it to make it more comfortable.
Pap test. A speculum is inserted into the vagina to widen it. Then, a brush is inserted into the vagina to collect cells from the cervix. The cells are checked under a microscope for signs of disease.
The technique involves the use of three different solutions, on the one side, one corresponding to hematoxylin, on the other hand, the one containing Orange G (Papanicolaou OG solution) and the last with the remaining dyes (Papanicolaou solution EA).
Composition of the Reagents of Papanicolaou Staining (Pap stain) Hematoxylin = 2.5g. Ethanol = 25ml. Potassium alum = 50g. Distilled water (50C) = 500ml. Mercuric oxide = 1-3g. Glacial acetic acid = 20ml.
The first staining solution contains haematoxylin which stains cell nuclei. Papanicolaou used Harriss hematoxylin in all three formulations of the stain he published. The second staining solution (designated OG-6), contains Orange G in 95% ethyl alcohol with a small amount of phosphotungstic acid.
The principle of pap stain Solution 1 has Hematoxylin stain, a basic nuclear stain. Solution 2 has OG-6, an acidic cytoplasmic stain, has orange G with phosphotungstic acid (6 denotes the used concentration of phosphotungstic acid). Orange G is an acid dye and stains keratin with a bright intense orange.
