Inorder to remove the standard field from view, Go to the fields which you want to hide and select there link and select the Set Field-Level Security button and uncheck the Visible checkbox for the profiles you do not want to see those particular fields.
You can of course set a field to both Read-Only and Mandatory, however the Read-Only part will win-out against it.
Salesforce Classic: Click on Setup | Manage Users | User. Open the user record and navigate to Permission Set Assignments section. Open the Permission Set assigned and go to System permission. Deselect the option Edit Read Only Fields Save.
Always On Layout fields are fields that must be on the page layout and cannot be removed from the Page Layout.
Salesforce Classic: Click on Setup | Manage Users | Permission Sets. Open the Permission Set that has the Edit Read-Only Fields added in it. Go to System permission. Deselect the option Edit Read Only Fields Save.
Open the field. Click Set Field-Level Security button. Select Read-Only check box for the profile.
All you have to do now is find the field youd like to hide and click, hold, and drag it up to the box of fields at the top of the page (so that you see the green ✅ appear), then release. And youre all set, the field is now hidden from the page layout!
Required and Read-only are generally mutually exclusive, and being able to blank out a field generally means that the field cant be required nor read-only (the exception being using a page layout to enforce these restrictions, as appears to be your case).
