Ideally, specimens should remain in fixative for long enough for the fixative to penetrate into every part of the tissue and then for an additional period to allow the chemical reactions of fixation to docHub equilibrium (fixation time). Generally, this will mean that the specimen should fix for between 6 and 24 hours.
Fixation of the specimen Kaiserling recommended that the initial fixation be a neutral formalin (KI) solution and then transferred to a final preserving glycerin solution (KIII) for long term display. Colour preservation is also maintained with these solutions.
Fixation consists of two steps: cessation of normal life functions in the tissue (killing) and stabilization of the structure of the tissue (preservation). The goal of fixation is to preserve structure as faithfully as possible compared to the living state.
The specimen is placed in a liquid fixing agent (fixative) such as formaldehyde solution (formalin). This will slowly penetrate the tissue causing chemical and physical changes that will harden and preserve the tissue and protect it against subsequent processing steps.
Gentle fixation for tissue arrays Place tissues in 4% paraformaldehyde for no longer than 48 hours at 4oC. The solution should completely cover the tissue.
Types of fixation Fixation of tissues can be achieved by chemical or physical means. Physical methods include heating, micro-waving and cryo-preservation (freeze drying). Heat fixation is rarely used on tissue specimens, its application being confined to smears of micro organisms.
