In the fields of histology, pathology, and cell biology, fixation is the preservation of biological tissues from decay due to autolysis or putrefaction. It terminates any ongoing biochemical reactions and may also increase the treated tissues mechanical strength or stability.
If possible, photograph and relax specimens before fixing. Put in a jar with enough seawater to allow the specimens or expand fully, then freeze or adds menthol or magnesium chloride. Leave until relaxed, fix in formalin for a maximum of 12 hours; rinse thoroughly in water, store in 70% alcohol.
Fixation is the foundation step behind the study of pathology and essentially exists to prevent the autolysis and degradation of the tissue and tissue components such that they can be observed both anatomically and microscopically following sectioning.
DEFINITION OF FIXATION AND STAINING Fixation may be defined as the selective preservation of cell or tissue structures and components for subsequent study. Fixation is selective in the sense that those methods are selected which are most suitable for the particular purpose of the research study.
Fixation The specimen is placed in a liquid fixing agent (fixative) such as formaldehyde solution (formalin). This will slowly penetrate the tissue causing chemical and physical changes that will harden and preserve the tissue and protect it against subsequent processing steps.
A sample is fixed in the field by adding chemicals that prevent the water quality indicators of interest in the sample from changing before final measurements are performed later in the laboratory.
Glutaraldehyde is primarily used to fix samples for electron microscopy (EM). It is often diluted to 2-4% in sodium cacodylate buffer just before use. Fixation with glutaraldehyde is strong and irreversible. Fixation with glutaraldehyde will dramatically change the conformation of many antigenic epitopes.
