The Papanicolaou stain is undoubtedly the most commonly used in cytology. It has three cytoplasmic dyes, orange G, eosin Y, and light blue. It can stain cells blue or blue-green (basophilia, cyanophilia), pink (acidophilia, eosinophilia), orange (orangeophilia), or indeterminate (gray-blue).
A procedure in which a small brush is used to gently remove cells from the surface of the cervix and the area around it so they can be checked under a microscope for cervical cancer or cell changes that may lead to cervical cancer. A Pap smear may also help find other conditions, such as infections or inflammation.
Acetic acid or vinegar is placed on the cervix and vagina to stain the cells and to allow the clinician to better see where the abnormal cells are located and the size of any abnormal areas. The size, type, and location of abnormal cells help to determine which area or areas may need to be biopsied.
EA 65 reagent, Pap 3C reagent is an alcoholic solution of two acid dyes, Eosin Y and Light Green SF, with added Bismarck Brown dye and phosphotungstic acid (PTA).
The first staining solution contains haematoxylin which stains cell nuclei. Papanicolaou used Harriss hematoxylin in all three formulations of the stain he published. The second staining solution (designated OG-6), contains Orange G in 95% ethyl alcohol with a small amount of phosphotungstic acid.
PAP bluing solution is a variant of Scotts solution and is used specifically in Papanicolaou staining (Pap staining) for cytological smears. It is used to bluish the hematoxylin in the stained nuclei to improve contrast and visibility, which facilitates the interpretation and analysis of cytology specimens.
Papanicolaou stain (also Papanicolaous stain and Pap stain) is a multichromatic (multicolored) cytological staining technique. The Papanicolaou stain is one of the most widely used stains in cytology, where it is used to aid pathologists in making a diagnosis.
Your healthcare provider will use a small brush, swab or spatula to gently remove cells from the cervix and back of the vagina. They will place the cells in a vial of liquid or smear the cells on a glass microscope slide. If you need an HPV test, your provider will take a sample of cells for this test as well.
