The overall goal of this video is to demonstrate an immunohistochemistry protocol for paraffin-embedded tissue sections. This is accomplished by first slicing paraffin infiltrated tissue on a microtome and then mounting the sections onto charged slides. The sections are then deparaffinized and rehydrated before performing antigen unmasking to remove the cross-links formed by fixation. Next, a specific antibody is applied to the sections to bind to its target epitope prior to chromogenic staining. After dehydrating the sections, the final step is to mount the sections for microscopic analysis. Ultimately, antibodies are used to detect and analyze protein expression while maintaining the composition, cellular characteristics, and structure of native tissue. For dried stains, pretreat or soak in warm water with a product containing enzymes, then launder. These types of laundry products are often recommended for protein-based stains, such as blood. Check the labels for this ingredient, as most enzyme products will promote their ability to remove these stains.
But if you are still wondering if one technique is better than another, or what it would take to start using a new sectioning method, keep reading. Well cover that here. The three primary means of embedding tissue for sectioning are paraffin wax, Optimal Cutting Temperature (OCT), and resin.
Lemon juice, known for its natural bleaching properties, can be applied directly to the stains or mixed with baking soda to form a paste. Allow the mixture to sit on the fabric for a few hours before rinsing it off. Sunlight is another ally in stain removal.
To clean mud off the exterior, use a high pressure hose water or a pressure washer to remove the caked on mud. Next, use a Foam Cannon or a foam gun to apply The Last Soap. Then let the vehicle settle for a few minutes. Once youve finished, use a Foam Cannon to clean the exterior of the car, this time in more detail.
Soak the item of clothing in warm water, then apply a few drops of dish soap on the stain. Hold the material around the stain and start rubbing the sides against each other to work up a lather. This will help the fabric absorb the soap. Let it sit for about an hour or so before putting it through the washing machine.
Embedding is important in preserving tissue morphology and giving the tissue support during sectioning. Some epitopes may not survive harsh fixation or embedding. The tissue is typically cut into thin sections (5-10 m) or smaller pieces (for whole mount studies) to facilitate further study.
Use a solution of 50 percent vinegar and 50 percent water on the stain. Apply liberally and then treat with laundry soap and water. DO NOT put the garment in the dryer until you have seen that the stain is gone, he warns.
Baking soda Try using it to create a stain-removing paste by mixing a tablespoon with water, or alternatively mix it with hydrogen peroxide or white vinegar. As well as spot-cleaning, you can also add baking soda to a regular wash or soaking clothes in water with baking soda overnight.
