For light microscopic examinations, colored agents (chromophores) are used. For the electron microscope, electron-dense heavy metal salts are employed (e.g., lead citrate, uranyl acetate). The earliest histologists used naturally occurring dyes to stain their sections.
HE. HE is the most commonly used of all the various staining methods available in frozen section. HE is simple to perform, inexpensive and reliable. The two main dye components are hematoxylin and eosin.
Positive staining stains the virus itself such that the virus is dark against a lighter background (Figure 1A). This is the easiest staining to do and yields results suitable for analyzing the morphological diversity of viruses from aquatic samples.
Thus, commonly used basic dyes such as basic fuchsin, crystal violet, malachite green, methylene blue, and safranin typically serve as positive stains. On the other hand, the negatively charged chromophores in acidic dyes are repelled by negatively charged cell walls, making them negative stains.
Embed skin diagonally, facing such a way that the microtome blade cuts through the fat and dermis first and epidermis/hair last. Arrange tissue ing to size in the mold, from smallest to largest, with the epidermis facing the same direction in all skin sections.
Tissue embedding is the process of placing tissues in molten paraffin before sectioning, using methods like metal molds or pans, to facilitate rapid embedding and ensure proper orientation for accurate morphology demonstration in samples.
The most widely used stains in electron microscopy are the heavy metals, uranium and lead. The double contrast method of ultrathin sections with uranyl acetate (UA) and lead citrate is the standard contrasting technique for electron microscopy (Figure 1 and 2).
TEM relies on negative staining of purified virus particles with an electron-dense material, such as uranyl acetate or phosphotungstate. An image is formed from the interaction of the electrons transmitted through the specimen; the image is magnified and focused onto a layer of photographic film (Fig. 2.8).
