Hematoxylin and Eosin Stain (or HE Stain) Hematoxylin is the most commonly used nuclear stain in histology and pathology although, despite its long use and honorable history, the chemistry of the dye is still not fully understood.
For example, one of the most common stains, Hematoxylin, is a basic dye that stains proteins a blue color, while Eosin stains proteins a pink color. These two stains are commonly used together to define intracellular organelles and proteins.
Embedding converts the tissue into a solid form which can be sliced (sectioned). Sectioning (slicing) provides the very thin specimens needed for microscopy. Staining provides visual contrast and may facilitate identification of specific tissue components.
Toluidine Blue O: Stain utilized for optimal demonstration of mineralized bone and osteoid seams, osteoblasts, osteoclasts.
Prussian blue: stains for iron content in bone tissue specimens. Elastin/Van Gieson (EVG): stain used to determine the presence of elastic fibers. Calcein: labels bone mineralization fronts in vivo. Oil red O: stain used to visualize the presence of adipose cells in bone specimens.
