PAP bluing solution is a variant of Scotts solution and is used specifically in Papanicolaou staining (Pap staining) for cytological smears. It is used to bluish the hematoxylin in the stained nuclei to improve contrast and visibility, which facilitates the interpretation and analysis of cytology specimens.
This bluing method consists in immersing the metal in boiling water with the addition of substances such as copper sulphate, lead acetate and sodium hyposulphite. Over time, the colour takes on different shades, from blue to grey to black. Once extracted from the water, the object is left to cool to room temperature.
BLUING REAGENT Buffered lithium carbonate-based reagent for converting red stained nuclei to blue. Bluing reagent is used for bluing the samples after staining with hematoxylin. It is also used in both regressive and progressive methods of HE staining, one of the most commonly used staining methods.
Procedure: Remove cover slip by incubation in xylene for a few hours to several days (until the coverslip can be removed easily). Rehydrate slides in 100%, 85%, and 75% ethanol, each for 30 sec at room temperature. Wash slides in deionized or distilled water for 5 min at room temperature.
Use fabric dye or fabric markers: If the bleach stain remains noticeable, you can consider using fabric dye or fabric markers to color over the bleached area. Choose a dye or marker that closely matches the original color of the garment and carefully apply it ing to the product instructions.
